Ảnh hưởng của tiền xử lý bằng enzyme carbohydrase đến hiệu suất trích ly protein từ bột hạt sen tách béo

Nghiên cứu này đánh giá hiệu quả tiền xử lý bằng 3 loại enzyme gồm α-amylase, cellulase và xylanase nhằm tối ưu hóa quá trình trích ly protein từ bột hạt sen tách béo. Trong thí nghiệm đầu tiên (thực hiện ở pH 5,0, 50°C, khuấy 200 vòng/phút và hòa tan protein ở pH 8,5), α-amylase liều lượng 1.000 U/g được lựa chọn nhờ đạt hiệu suất trích ly cao nhất (47,90%), vượt trội hơn cellulase (39,60%) và xylanase (36,70%). Tiếp tục khảo sát ở thí nghiệm 2, kết quả cho thấy cả tỷ lệ nguyên liệu:nước (1:10-1:30 w/v) và thời gian xử lý (1,5-4,5 giờ) đều ảnh hưởng có ý nghĩa đến hiệu suất (p < 0,05), dù không có sự tương tác qua lại giữa hai yếu tố này (p = 0,287). Điều kiện tiền xử lý tối ưu được xác định là tỷ lệ 1:20 (w/v) trong 3 giờ, giúp nâng hiệu suất trích ly protein lên 56,90%. Kết luận cho thấy, α-amylase là enzyme phù hợp nhất để phá vỡ cấu trúc ma trận giàu tinh bột của hạt sen, giúp quá trình thu hồi protein đạt hiệu quả cao và tiết kiệm tài nguyên.
