Effects of extraction conditions on the antioxidant and antibacterial activities of hydroponic pennywort tuber extracts (Centella asiatica (L.) Urban)

Abstract

This study investigates the effects of three extraction parameters: solid-to-solvent ratio (1:10, 1:20, and 1:30 g/mL), extraction temperature (40–80°C), and extraction time (40 - 70 min), on the antioxidant activity and antibacterial activity of hydroponic pennywort tuber extracts against Escherichia coli and Staphylococcus aureus. The optimal conditions were a solid-to-solvent ratio of 1:20, 70°C, and 60 min, producing DPPH antioxidant activity of 15.42 ± 0.19 µmol TE/g. At 1,000 mg/mL, the extract produced inhibition zones of 16.00 ± 1.00 mm against E. coli and 18.33 ± 0.58 mm against S. aureus, comparable to ampicillin at 100 µg/mL. These findings provide a scientific basis for developing efficient extraction processes for functional food and medicinal applications.
